Bioluminescence: mechanism and mode of control of scintillon activity.

چکیده
Subcellular particles, termed scintillons, isolated from the luminescent dinoflagellate Gonyaulax, emit light when the pH is lowered from 8 to 5.7. The capacity to emit a second flash is regained by incubation of scintillons with the low molecular weight luciferin from Gonyaulax. This compound was previously demonstrated to be the substrate in the bioluminescent reaction catalyzed by a soluble luciferase isolated from the same cell. A model of the scintillon is presented that involves soluble elements structured within a membrane-bound particle.

The Characterization of Scintillons Bioluminescent particles from the marine dinoflagellate, Gonyaulax polyedra
A new type of biological particle, isolated from the marine dinoflagellate Gonyaulax polyedra, has been partially purified and characterized. When the pH is lowered, the particle emits light in vitro in a fashion closely mimicking the flash of the living cell, and it is referred to as a scintillon (flashing unit). Scintillons are obtained by breaking the cells in buffer at pH 8.2 and purifying ...
متن کاملIdentification of a vacuolar proton channel that triggers the bioluminescent flash in dinoflagellates
In 1972, J. Woodland Hastings and colleagues predicted the existence of a proton selective channel (HV1) that opens in response to depolarizing voltage across the vacuole membrane of bioluminescent dinoflagellates and conducts protons into specialized luminescence compartments (scintillons), thereby causing a pH drop that triggers light emission. HV1 channels were subsequently identified and de...
متن کاملDirected Improvement of i-Photina Bioluminescence Properties, an Efficient Calcium-Regulated Photoprotein
Photoproteins are excellent reporter systems because they don’t have virtually background signal. Aequorin is the most well-known photoprotein. Three improved engineered photoproteins photina, i-photina and c-photina, were also recently developed and optimized for generation of Ca2+ mobilization assays precisely. The total light emission is greater than aequorin and their reacti...
متن کاملSensorless Indirect Field Oriented Control of Single-sided Linear Induction Motor With a Novel Sliding Mode MRAS Speed Estimator
This paper proposes a new sliding mode control (SMC) based model reference adaptive system (MRAS) for sensorless indirect field oriented control (IFOC) of a single-sided linear induction motor (SLIM). The operation of MRAS speed estimators dramatically depends on adaptation mechanism. Fixed-gain PI controller is conventionally used for this purpose which may fail to estimate the speed correctl...
